ADC Therapeutics Announces Completion of Enrollment in LOTIS-7 Phase 1b ZYNLONTA® Combination Trial

ADC Therapeutics completed LOTIS-7 enrollment with 100 relapsed/refractory DLBCL patients dosed at the selected ZYNLONTA 150 µg/kg plus glofitamab regimen across 30 sites (70% US/30% EU), with full data expected in Q4 2026. Prior LOTIS-7 efficacy data showed an 89.8% overall response rate and 77.6% complete response on 49 efficacy-evaluable patients, with a manageable safety profile. The company plans to present full results at a medical meeting and submit for publication by end-2026, supporting a continued push toward “best-in-class” 2L+ bispecific combination potential.

Analysis

This is more de-risking than de-risked value creation. Completion of enrollment only increases the probability that the combo can be validated later; it does not change near-term revenue, and for a balance-sheet-sensitive microcap like ADCT the stock still trades primarily on financing/approval optionality. The market should be careful not to capitalize a best-in-class claim until durability and infection burden are visible in a materially larger dataset.

The real second-order effect is strategic, not commercial: pairing with Roche’s glofitamab could make ADCT a useful complement inside an established bispecific franchise, but that also means Roche likely captures bargaining power if the regimen works. If efficacy holds, the winners are likely Roche and ADCT’s development optionality; the losers are alternative 2L+ DLBCL regimens that depend on simpler administration or cleaner tolerability, especially rival bispecific programs from ABBV/Genmab and other ADC combinations that have to defend against a stronger CD19/bispecific stack.

Catalyst timing matters. Over the next 1-3 months, this should be a low-signal headline unless management uses it to raise capital or tease earlier readouts. The real inflection is Q4 2026 full data; by then, the key falsifiers are any slip in response durability, a materially worse safety profile after mandatory prophylaxis is incorporated, or an FDA view that the combo remains exploratory rather than pathway-enabling. If that happens, today’s optimism likely compresses back to a financing overhang trade.
