Truist raised its Septerna (SEPN) price target to $43 from $35 while keeping a Buy rating, implying ~28% upside from $33.49; the stock is up 216% over the past year. The model update increased the probability of success for its hypoparathyroidism program to 35% from 20%, with Phase 1 healthy volunteer readout expected in late 2026 or the first half of 2027, while Truist assigns no value to the SEP-631 program after a competitor’s MRGPRX2 miss. The incremental information is less about today’s target raise and more about how the market is pricing a very long-dated platform story: SEPN is now trading on scarcity value of a de-risked GPCR engine, not on any near-term cash-flow or even near-term clinical inflection. That matters because the stock can stay expensive for months on analyst enthusiasm, but the tape is vulnerable if the market starts discounting the 2026/27 readout horizon more heavily than the platform optionality.
